internal standard for peptide digest
Add 10 uL of glu-fib 500 fmol/uL for 100 uL of sample (at 100fmol/uL). Works fine.

start easy-nanosprayer on maXis
1. Replace the normal spacer with "Low Flow spacer".
2. Check the tip of the sprayer if the core capillary is fully retracted in the steel tube.
3. Insert the nanospray assembly on to the ESI source.
4. Turn the gold wheel clockwise to extend the core capillary (can't observe).
5. Connect syringe pump with tune mix (L ESI-TOF version) flow at 60 uL/h)
6. Adjust nebulizer as required.
The easy-nanospray needs to be primed with 1uL/min flow of mobile phase or tune mix solution. I prefer the latter, as I can check and recalibrate the system at the same time. At 1 uL/min flow, nebulizer was set as low as 0.1 bar to achieve the most intense signal.

Integration of Shimadzu Prominence HPLC to HyStar
1. Back up system
2. Connect Network Card #1 to CBM-20A Lite (with or without Ethernet Switch)
3. Setup a static IP address 10.104.52.20 for Network card #1
4. Install the Shimadzu-HyStar plug-in
5. In HyStar's hardware setup, choose Shimadzu HPLC as the LC System
6. Click LC System Settings
7. Choose CBM-20A Lite as controller, click "configure"
7. Specify IP address of 10.104.52.14 for CBM-20A Lite Connection
8. Configure other devices (except Detector--which should be left blank)
9. Click Save
10. In HyStar hardware setup, click close.

How to make Dell Latitude E6410 dual boot
My E6410 is pre-installed with Windows 7 professional but my software required windows XP to run. The following steps were necessary for making it dual boot.
1. Resize original Windows 7 partition to 40G (a C: partition) and create a new ntfs primary partition (a D: partition) using Gparted
2. Install windows XP professional. You can get an old winXP pro CD you have, borrow it or download it.
3. With Windows XP installed on D: drive, download easyBCD AND dotnetfx 2.0 Install dotnetfx 2.0 first then easyBCD.
4. Run easy and Add New Entry, and select "Windows 7" as type and C:\ as drive. Finally click Add Entry.
5. In Bootloader setup choose "Install widows 7 bootloader on MBR" and click "write MBR".
6. Exit easyBCD and reboot in to windows 7.
7. copy the make and model of hardware (network card, video, wirteless, etc using device manager). Then download at least the wireless and network. As of Nov 26, 2010, the network card is Intel 82577LM Gigabit Network

Preparation of 1ppm aspartam solution for MTQ injection
1 sachet of Equal contains 36mg of aspartam.
1 sacket in 500mL of water gives 36mg/600mL concentration = 0.06 mg/mL = 60ppm in water.
Add 10uL of this solution to 590 uL of water gives 1ppm solution of aspartam.

How to update ncbi database on MASCOT
Download ncbi nr database from the following link. Download the nr.gz file. Once done, take the following steps.
1. Extract the nr.gz. Copy the file to D:\NETPUB\mascot\sequence\ncbinr\incoming. Rename the extracted file to ncbinr_YYYYMMDD.fasta, where YYYYMMDD is the current date.
2. On database maintenance page, select ncbinr and set it to "inactive". Click OK.
3. Move the files in D:\NETPUB\mascot\sequence\ncbinr\current to D:\NETPUB\mascot\sequence\ncbinr\old. Then, move the file in 1 to D:\NETPUB\mascot\sequence\ncbinr\current.
4. Finally, wait until the file is indexed, go back to database maintenance page and set the status of ncbinr database to "active".
