Workflow for Quantitation on MTQ

Things to ptimize transfer, then ionization and finally calibration.

1. Prepare 1 ppm solution of analyte standard in 50:50 Water:MeOH or ACN.
2. Infuse and optimize parameter to maximize signals
CC RF, Q lowmass, Transfer time (add EIC of compound of interest). (flow rate 180-240 uL/hr)
3. With a Tee, connect HPLC flowing with mobile phase at the desired flow rate. Optimize nebulizer gas and dry gas.
4. Add calibration segment and adjust the run time.
5. Remove the Tee and infuse calibrant and calibrate the instrument.
6. Save method as.

Note. Changing transfer time to maximize signal may shorten the mass range. The calibration mode may have to be adjusted. (for Caffeine, we used NaFormate and had to use TOF calibration version 2 (with Enhanced Quadratic as calibration mode) because the mass range only extends from 158 to 400 and only a few calibration points are visible.
